Wendy Naimark, PhD, VP R&D, introduced herself as a veteran of cardiovascular local drug delivery and new to ophthalmology. About 2 years ago Interface made a discovery, a way to engineer novel non-polymeric prodrugs with chemistry and processing to create standalone controlled release implants. The technology strengths include sustained zero-order release kinetics, drug release controlled via surface erosion, drug release concurrent with the erosion of the implant, the no polymer matrix allows a high drug ratio and smaller implant design, and polymer-like properties provide flexibility for product design. The prodrugs can be used to create a coating, and solid forms such as a cylinder and nanoparticles. Interface is starting out with steroids, for use in treating posterior inflammation (DME, RVO, non-infectious uveitis) and following cataract surgery, and moving into non-steroid programs in wet AMD and glaucoma. The company’s lead program is the IBE-814 intravitreal implant, which is a dexamethasone prodrug cylinder. A trial in rabbits showed near-zero drug release in the vitreous humor for at least 6 months, and the implant demonstrated sustained inhibition of VEGF-induced vascular leakage for at least 6 months.
